Glycyl-leucyl-methionine (Gly-Leu-Met-NH₂) is the 9- 11 residue peptide of the neuropeptide Substance P. Conformational studies have shown that the tripeptide has a twist-like structure and can weakly bind to the neurokinin-1 (NK1) receptor, which helps the study of receptor binding kinetics and structure-activity relationship. In vascular smooth muscle experiments, SP(9–11) exhibited vasodilation and spasm-inducing effects at high concentrations, showing that longer SP fragments have lower agonist potency. Although the activity is far inferior to that of the complete Substance P, SP(9–11) is still an important molecular probe for analyzing the NK1 receptor activation pharmacophore and peptide design.
